Objective
Yiqifumai injection (YQFM) has a clinical advantage in the treatment of shock‐related diseases. However, whether YQFM can improve pulmonary microvascular injury during hemorrhagic shock and resuscitation (HS/R) and the mechanisms remain unclear.
Methods
Rats HS/R model was established by withdrawing arterial blood and reinfusion. YQFM was infused intravenously for 1 h immediately after resuscitation. Vital sign examination, histology, immunohistochemistry and immunofluorescence, flow cytometry, enzyme‐linked immunosorbent assay, and Western blot analysis were utilized to investigate the attenuating effect and mechanisms influenced by YQFM.
Results
YQFM inhibited the decrease of rat survival rate and improved animal vital sign after HS/R. Meanwhile, YQFM attenuated HS/R‐induced increase in leukocyte infiltration into lung tissues, Evans blue extravasation out of pulmonary microvessels and lung edema, along with a downregulated expression of junction proteins. Besides, HS/R‐induced a decrease of basement membrane protein and an increase of matrix metalloproteinases expression and Cathepsin B cleavage, which were all attenuated by YQFM. Moreover, YQFM inhibited the decreased ATP/ADP and ATP/AMP ratio, as well as mitochondrial complexes activity and low expression of ATP synthase subunit δ (ATP5D) after HS/R.
Conclusions
YQFM attenuated HS/R‐induced pulmonary microvascular barrier damage through both improving energy metabolism mediated junction integrity and ameliorating basement membrane degradation.
