Siglec-15 and PD-1 checkpoint blockade in combination with oncolytic Zika virus infection confers protection against immune-resistant gliomas

Lay Summary

Siglec-15 is a protein that is expressed on glioblastoma cells and immune cells. Using fresh human tumor specimens, we identified which specific immune cells express Siglec-15. Patients whose tumors express more Siglec-15 live shorter, on average. Blocking Siglec-15 activated immune cells to kill tumor cells. Combining its blockade with a cancer-targeting virus, Zika virus, and an immunotherapy targeting PD1, mice with brain tumors lived significantly longer. These findings suggest combination therapy with blockade of Siglec-15 might be helpful for patients.
