Background

Periodontitis is an inflammatory disease that damages tooth-supporting structures leading to tooth loss. Zoledronic acid (ZA), a potent antiresorptive agent, has recently shown promise for dental regenerative applications.

Aim

To evaluate the potential regenerative impact of topical ZA gel on ligature induced periodontitis in rats.

Materials and methods

Thirty male albino rats were assigned into three groups. Groups II and III exposed to periodontitis induction for 2 weeks. Group I (negative control, n = 6), group II (positive control, n = 12) was given daily topical saline for 2 weeks, group III (ZA, n = 12) was given topical single dose of ZA 16 μg/100 μl. Two and 4 weeks post-ligature removal and treatment beginning, half of the rats in each group were euthanized then specimens were examined by light and scanning electron microscopes.

Results

Group II showed the periodontal apparatus with marked destruction and a highly significant increase in the alveolar bone loss ( P < 0.0001). Group III showed relative regeneration and a significant decrease of alveolar crest level compared to group I at 2 weeks ( P = 0.002). At 4 weeks, group III showed markedly regenerated periodontal apparatus ( P = 0.001).

Conclusion

ZA gel significantly promoted tissue regeneration in albino rats with ligature induced periodontitis.
