Real-world adherence to and persistence with ofatumumab, infusion, oral, and other self-injectable disease-modifying therapies in patients with multiple sclerosis

Background

Treatment schedule complexity and route of administration may influence patient adherence and persistence. Limited real-world evidence exists comparing these outcomes across treatments for multiple sclerosis (MS).

Objective

To examine 18- and 24-month adherence/persistence of ofatumumab (OMB) and other approved disease-modifying therapies (DMTs).

Methods

Optum Clinformatics® Data Mart data were used to identify adult patients with MS who initiated treatment with an approved DMT from August 20, 2019, to December 31, 2023. Patients were stratified into four treatment groups: OMB, infusion DMTs, oral DMTs, and other injectable DMTs.

Results

After propensity score matching, adherence to OMB was significantly higher than to oral DMTs (18 months: 73% vs. 51% [ p  < .001]; 24 months: 69% vs. 47% [ p  < .001]) and was also higher with OMB than with other injectable agents (18 months: 74% vs. 40% [ p  < .001]; 24 months: 70% vs. 35% [ p  < .001]). Adherence to OMB and infusion DMTs was comparable (18 months: 74% vs. 70% [ p = . 31]; 24 months: 70% vs. 65% [ p = . 24]). Similar patterns were observed for persistence between the groups.

Conclusion

These findings suggest OMB is a convenient, effective, self-administered alternative to infusion therapies.
