One-Pot Aqueous-mediated Synthesis of 2,5-disubstituted 1,3,4- thiadiazoles via Iodine-Promoted Oxidative Cyclization

A practical and transition-metal-free protocol has been developed for the synthesis of 2,5- disubstituted 1,3,4-thiadiazoles via iodine-promoted oxidative cyclization of benzothiohydrazides with aldehydes in aqueous medium. The methodology proceeds via efficient intramolecular C–S and C–N bond formation under mild conditions, using K₂CO₃ as the base and water as the solvent. A broad range of substituted aromatic aldehydes bearing electron-donating and electron-withdrawing groups were well tolerated, affording the corresponding thiadiazole derivatives in good to excellent yields.
