The enzyme Ilß-hydroxysteroid dehydrogenase (11ß-HSD2) provides mineralocorticoid receptor specificity for aldosterone by metabolizing glucocorticoids to their receptor inactive Il-dehydro derivatives. Glycyrrhizic acid (GA), a recognized inhibitor of 11ß-HSD2, is a component of liquorice. We recently provided first evidence that aldosterone receptor antagonism normalizes blood pressure, prevents up-regulation of vascular ET-l and restores nitric oxide-mediated endothelial dysfunction, and may thus advance as a novel and specific therapeutic approach in 11ß-HSD2-deficient hypertension..
