Fractionation of Ocimum sanctum (Tulsi) Extract and Evaluation of Its Antimicrobial, Antioxidant and Anti-Inflammatory Properties with Potential Applications in Periodontal Wound Healing – An In Vitro Study

Abstract

Background:

Periodontal disease is a chronic inflammatory disorder, perpetuated by microbes and maintained by inflammatory responses of the host. Pleiotropic effects of herbal bioactives have immense scope as adjuncts in periodontal therapy.

Objective:

Assessment of the antimicrobial, anti-inflammatory, and antioxidant properties of fractionated bioactives from Ocimum sanctum (Tulsi) using in vitro methods.

Methods:

Tulsi leaves were extracted using five solvents of increasing polarity. The most active extract was fractionated by column chromatography, and three bioactive fractions were combined (1:1:1) to prepare a paste. Antibacterial, antioxidant, and anti-inflammatory activities were evaluated using agar well diffusion, 2,2-diphenyl-1-picrylhydrazyl (DPPH), and human red blood cell (HRBC) membrane stabilisation assays. Experiments were performed in triplicate, and data are presented as mean ± standard deviation (SD).

Results:

Tulsi fraction paste showed antibacterial activity against the tested periodontal pathogens, with greater efficacy against P. gingivalis . It also demonstrated dose-dependent antioxidant and anti-inflammatory activities in the DPPH and HRBC membrane stabilization assays.

Conclusion:

Fractionated bioactives of Ocimum sanctum exhibited antioxidant, antimicrobial, and anti-inflammatory properties in vitro . The results suggest potential therapeutic applications in wound healing and tissue repair, which could be further explored in vivo and in clinical settings.
