Evaluation of the Effectiveness of Dilution Methods in CBC Analyses in Lipemic Samples

Objectives: Studies evaluating procedures to reduce lipemia interference in complete blood count (CBC) testing are limited. The aim of this study was to evaluate CBC results in lipemic samples after treatment with two different lipemia-removal methods.Methods: Blood samples from patients with hypertriglyceridemia were used in this study. Sixty patients with hypertriglyceridemia who had two K2-EDTA whole-blood tubes collected simultaneously were included. Two lipemia-removal procedures were evaluated: Plasma replacement with saline and whole-blood dilution with saline. CBC parameters measured on the UniCel DxH 800 analyzer were compared between the treated samples and the original lipemic samples.Results: The WBC, RBC, and platelet counts were significantly lower following both procedures compared with the original lipemic samples. Hemoglobin, MCH, and MCHC values were significantly lower after plasma replacement with saline, whereas no significant changes were observed after whole-blood dilution compared with the original lipemic samples.Conclusion: Plasma replacement with saline appears to be more effective than whole-blood dilution for reducing lipemia-related interference in CBC measurements and may be considered for routine laboratory practice.
