ABSTRACT

Non‐small cell lung cancer (NSCLC) is among the leading causes of cancer‐related deaths worldwide. Previous treatment regimens often have limited efficacy due to drug resistance and recurrence. Metabolic reprogramming participates in the initiation and progression of NSCLC, with prominent changes in lipid metabolism. Mutations in specific driver genes directly remodel lipid metabolic reprogramming in NSCLC by rewiring lipid synthesis, uptake, and catabolism, which confers cancer cells with enhanced proliferative and metastatic capacity. On this basis, targeted lipid metabolism reprogramming has become a promising new direction for the treatment of NSCLC. In this review, we examined the alterations in lipid metabolism in NSCLC, explored the mechanisms by which driver gene mutations change lipid metabolism in NSCLC, and provided possible therapeutic strategies targeting key lipid metabolism pathways at the current stage, aiming to offer a theoretical basis and prospects for the development of novel therapies targeting specific gene mutation subtypes of NSCLC.
