Direct STAT6 Inhibition: A Small-Molecule Strategy for Intercepting Type 2 Inflammation at Its Transcriptional Core

Anna C. Renner, Robert B. Kargbo

Abstract

STAT6 integrates interleukin-4 and interleukin-13 signaling into transcriptional programs that drive type 2 inflammation. A new chemotype of heterocyclic biphenyl-3-yl carboxamides directly targets this signaling node. By moving downstream of cytokine receptors while avoiding broad upstream JAK blockade, these molecules suggest an oral strategy for suppressing allergic disease with potentially greater pathway precision, although pharmacological validation remains essential.
