Development and Optimization of Transdermal Patch of Methimazole for the Treatment of Hyperthyroidism

Abstract

Background:

Antithyroid medications like methimazole are frequently used to treat hyperthyroidism, which is defined by an excess of thyroid hormone production. However, because of its short half-life, methimazole must be taken often by mouth, which might result in variable plasma concentrations and decreased patient compliance.

Aim and Objective:

In order to offer prolonged drug release, increase therapeutic effectiveness, and improve patient adherence, this study set out to design and optimize a transdermal patch formulation of methimazole.

Materials and Methods:

Methimazole and the excipients (pullulan, chitosan, polyvinyl alcohol, Transcutol, propylene glycol, and glycerol) did not significantly interact, according to Fourier transform infrared spectroscopy. Using a central composite design, the formulation was adjusted using response variables for drug flux, tensile strength, and folding endurance.

Results:

With 82.88% of methimazole diffusing across the test period, in vitro drug release experiments showed a sustained release profile, indicating the patch’s potential for long-lasting therapeutic activity. In order to guarantee its longevity and flexibility, the transdermal patch demonstrated appropriate mechanical qualities, such as sufficient tensile strength and folding endurance.
