Development and Comprehensive Greenness Assessment of a ZIC‐HILIC Chromatography for the Determination of the Anticancer Drugs in Pharmaceutical Preparations

ABSTRACT

The objective of the research was to examine the chromatographic properties of two anticancer drugs, Brigatinib and Alectinib, in hydrophilic interaction liquid chromatography (HILIC). The anticancer drugs were separated and quantified using chromatographic methods on two Lab‐made HILIC columns (ZIC‐1 and ZIC‐4), with varied alkyl spacer lengths between the charged functional groups. The development of the method relied on the effect of separation parameters, including the percentage of acetonitrile, pH, and acetate buffer concentration, which influenced the retention factors of the chosen drugs. The optimum separation conditions were determined with the help of two columns manufactured in the lab (100 mm × 4.6 mm I.D., 3.5 µm particle size) whose eluent was composed of both 20% acetate buffer (50 mM, pH 4.75) and 80% acetonitrile with a flow rate of 0.6 mL/min. The separation was monitored using a UV detector at 260 nm. There are five well‐established greenness metrics for the chemical methods assessed. Among them, AES, AGREE, NAMI, BAGI, and GAPI have been identified as valuable and trustworthy assessment instruments.
