The present work reveals an optimized protocol for the synthesis of novel six‐membered 2‐amino‐1,3,4‐thiadiazine derivatives of N α ‐protected amino acids, using N α ‐Fmoc/Cbz‐amino acid‐derived bromoketone in conjunction with thiosemicarbazide. In silico studies highlighted promising interactions for the anticancer proteins EGFR and MMP‐9 of 10 compounds (4a–4j) synthesized. Compound 4e showed a significant docking score of −8.4 kcal/mol and compound 4j scored −7.9 kcal/mol for EGFR. Additionally, compound 4e exhibited a strong binding affinity of −9.2 kcal/mol, and compound 4j scored −8.7 kcal/mol for MMP‐9. Overall, the synthesized 2‐amino‐1,3,4‐thiadiazines (4a–4j) demonstrate encouraging interactions for potential cancer therapy.
