Comparative Effect of Pregabalin and Lavandula angustifolia on Behavioral Changes in Rats’ Model of Epilepsy

Abstract

Background:

Linalool is an aromatic monoterpene alcohol widely found in essential oils. Linalool is one of the main bioactive components of Lavandula angustifolia . Linalool has an inhibitory effect through gamma-aminobutyric acid receptors. This effect inhibits the limbic system and reduces anxiety.

Objectives:

Explore the potential anti-epileptic activity of L. angustifolia (linalool) on behavioral changes in a rat model of epilepsy.

Materials and Methods:

Twenty-five adult Wistar albino male rats, aged about 12 weeks old and weighing between 300 and 350 g were gathered and habituated in an animal house of the College of Medicine, Babylon University. Rats were divided into five groups as follows: Group 1: control group, Group 2: PTZ, Group 3: Pregabalin + PTZ. Group 4: Linalool + PTZ, Group 5: the combination group, Pregabalin + Linalool + PTZ.

Results:

In Groups 3: Pregabalin + PTZ (12.8 mg/kg + 40 mg/kg), Group 4 Linalool + PTZ (300 mg/kg + 40 mg/kg), Group 5: the combination group, Pregabalin + Linalool + PTZ (12.8 mg/kg + 300 mg/kg + 40 mg/kg). The results showed a significant increase in latency and decreases in duration and severity of epilepsy compared to group 2 (PTZ group).

Conclusion:

Behavioral changes (decreasing latency period, increasing severity and duration of fits) induced by pentylenetetrazole were found to be opposed by linalool, pregabalin, and a combination of linalool with pregabalin via attenuating the severity and duration of fits, and by increasing the latency period of fits in male rats.
