Background:

The attainment of specific treatment objectives remains elusive in diabetes management. In diabetes mellitus (DM), pioneering antidiabetic medications, which can address multiple facets of the condition concurrently is anticipated. With this background, the study was conducted to evaluate the effect of four Ayurvedic polyherbal formulations in streptozotocin (STZ)-induced DM in Wistar rats.

Materials and Methods:

Diabetes was induced through two intraperitoneal injections of STZ at a dose of 30 mg/kg body weight, administered 1 week apart, with the compound dissolved in 0.9% normal saline. Rats were divided into seven groups, consisting six animals each. Group 1 (normal control - NC received water; Group 2 (diabetic control); Group 3 (metformin 100 mg/kg b.wt,); Group 4 ( Varadi Kwatha 900 mg/kg b.wt.); Group 5 ( Mustadi Kwatha 900 mg/kg b.wt.); Group 6 ( Khadiradi Kwatha 900 mg/kg b.wt.); and Group 7 ( Bhunimbadi Churna 450 mg/kg b.wt). A volume of 1–2 ml was administered in Groups 4, 5, and 6 through oral gavaging twice a day. The duration of treatment was 28 days. Serum glucose, serum triglyceride, total cholesterol, high-density lipoproteins-cholesterol, serum creatinine, total protein, alanine transaminase, aspartate amino transferase, alkaline phosphatase), serum glutamic-oxaloacetic transaminase, and oral glucose tolerance test (OGTT) were assessed on day 0, day 14, and day 28. Histopathological analysis of the liver, pancreas, and kidney of two representative samples from each group was performed.

Results:

Highly significant reduction in blood glucose levels ( P < 0.001) was seen in Varadi Kwatha , Mustadi Kwatha , Khadiradi Kwatha, and Bhunimbadi Churna . The percentage reduction in the OGTT values was significant in Khadiradi Kwatha ( P < 0.001) and Bhunimbadi Churna ( P < 0.001). Khadiradi Kwatha showed a significant decrease ( P < 0.05) in triglycerides. The trial formulations ameliorated the alterative changes caused by STZ-induced diabetes in liver, kidney, and pancreas.

Conclusion:

Varadi Kwatha , Mustadi Kwatha , Khadiradi Kwatha, and Bhunimbadi Churna are effective in STZ-induced diabetes in Wistar rats, Khadiradi Kwatha and Bhunimbadi Churna having more effective, in comparison to Varadi Kwatha and Mustadi Kwatha.
