An Investigation of N-Acetylcysteine’s Possible Hepatic Protection in Carbon Tetrachloride-Induced Hepatotoxicity in Rat Models

Abstract

Background:

The liver plays a crucial role in detoxifying the body by metabolizing internal and external toxic chemicals in most vertebrates. These substances can cause liver damage during normal body processes. Liver injury is primarily attributed to several reasons, such as viral infections, medication irritations, chemical toxicants, alcohol-induced oxidative stress (OS), inflammation, and immunological reactions. A recent study has shown that liver damage is mainly driven by inflammation and OS The release of cellular enzymes into plasma, such as alanine transaminase (glutamic–pyruvic transaminase [GPT]; ALT), aspartate aminotransferase (glutamic–oxaloacetic transaminase [GOT]; AST), and alkaline phosphatase (ALP), is a clear indicator of liver damage.

Objective:

To analyze the effect of N -acetylcysteine (NAC) on the carbon tetrachloride (CCl4)-induced hepatotoxicity in male albino rats.

Materials and Methods:

The study included 30 adult male albino rats. With a certain commercial feed and water, these rats became accustomed to living in an animal house. Following 10 days of acclimatization, the chosen rats were randomly assigned to three groups. For every series of studies, 10 rats have been used. The study was conducted at the Animal House in the College of Medicine, University of Babylon, from December 18, 2023, until January 1, 2024. The ALT, glutathione, and tumor necrosis factor alpha (TNF- α ) were measured in blood samples that were obtained.

Results:

The findings indicated that NAC protected the liver in the treated group, which differed significantly from the positive control group (induction by CCl4). The treated group with NAC had lower ALT levels, a highly significant increase in GSH levels, and a highly significant decrease in TNF- α levels compared to the positive control group. These findings demonstrated the hepatoprotective effect of NAC in male albino rats.

Conclusion:

The results illustrate the liver protection properties of NAC.
