An Khúc Thị <p>Ganoderma neo-japonicum is a valuable medicinal mushroom with
significant potential for pharmaceutical and biotechnological applications.
However, optimal conditions for its mycelial cultivation remain limited. This
study evaluated the effects of incubation temperature (15, 20, 25, 30, and
35°C) and culture media on the mycelial growth of a G. neo-japonicum strain
collected from Khanh Hoa Province, Vietnam. Five media were compared: potato
dextrose agar (PDA), PDA supplemented with rice bran (PRB) or yeast extract (PYE) or mineral salts (PKM), and malt extract medium (ME). Colony
diameter, growth rate, and morphology were recorded over 14 days. Temperature
significantly affected mycelial growth, with active expansion occurring
exclusively at 25 and 30°C. Although 30°C promoted faster initial growth, 25°C
yielded more stable development, producing dense, uniformly white mycelia and
the largest final colony diameter. Prolonged incubation at 30°C resulted in
reduced mycelial density and yellowish pigmentation. Culture media also markedly
influenced development: PYE supported the fastest early expansion, PRB achieved
the greatest overall growth and largest final colony diameter, whereas ME
resulted in the slowest growth. In conclusion, incubation at 25°C on PDA
supplemented with rice bran provides the most suitable conditions for
laboratory cultivation of G. neo-japonicum, providing a reliable basis for
future spawn production and domestication.</p>
